چکیده

The concept of peptic ulceration and its treatment is based on the ulcerogenic action of acid and pepsin (Card, 1952). It appears that pepsin plays the predominant role in this action (Schiffrin, 1940; Schiffrin and Warren, 1942; Le Veen, 1947), the acid merely providing the correct pH for optimal peptic activity. Consequently, it is of some importance to know the effect ofpH on the peptic activity of pepsin. One of these effects, the effect of pH on peptic activity, has been adequately studied (Christensen, 1955; Taylor, 1959a, b and c); the effect ofpH on enzyme stability has received little attention apart from the observations of Langley in the last century (Langley, 1881a and b) and those of Northrop, Kunitz, and Herriott (1948). As both of these effects are of considerable importance in clinical gastroenterology, the present study was undertaken of the effect ofpH on the stability and activity of pepsin.

Metabolism and Nutrition: Phytase and Minerals

An experiment was carried out to evaluate effects of temperature, pH, and incubation time on the stability of intrinsic phytases. The experiment was conducted as a 3*4*3*3 factorial arrangement with 3 feed ingredients (rye, wheat, and barley), 4 temperatures (T) (20, 38, 55, and 80°C), 3 pH levels (3, 5.5, and 8), and 3 incubation times (30, 60, and 120 min), with 2 replicates per treatment. Af...

Ovalbumin digestion by human pepsins 1, 3 and 5.

1. Of the three major human pepsins, pepsin 1 has greater proteolytic activity towards ovalbumin than has pepsin 3. Pepsin 5 has low activity towards this substrate. 2. Proteolytic pH-activity curves show only on pH maximum, about pH 1.4 for pepsin 1, pH 1.4--1.5 for pepsin 3 and pH 1.2--1.4 for pepsin 5. The curve for pepsin 3 has a shoulder between pH 2.4 and 3.4. 3. The rate of digestion of ...

Study on Activity and Stability of Proteases from Bacillus Sp. Produced by Submerged Fermentation

Objective: Investigations were carried out to isolate bacteria from saline-alkali soils and determined optimized alkaline protease activity and stability produced by a wild strain of bacillus sp. in submerged fermentation (SMF). Methods: Optimum temperature for enzyme activity in the crude extract was 40 ◦C at a pH between 8.0 and 9.0. The studies on pH stability showed that the enzyme...

Activity/stability of human pepsin: implications for reflux attributed laryngeal disease.

OBJECTIVES/HYPOTHESIS Exposure of laryngeal epithelia to pepsin during extra-esophageal reflux causes depletion of laryngeal protective proteins, carbonic anhydrase isoenzyme III (CAIII), and squamous epithelial stress protein Sep70. The first objective of this study was to determine whether pepsin has to be enzymatically active to deplete these proteins. The second objective was to investigate...
